Kurzfassung auf Deutsch: In der vorliegenden Arbeit konnten der Aufklärung des Biosyntheseweges des Sesquiterpen-Metabolismus der Sonnenblume (Helianthus annuus) entscheidende Schritte hinzugefügt werden. Hierzu wurden zunächst aus einer Transkriptom Datenbank Kandidaten Sequenzen anhand von Expressions-Mustern und Ähnlichkeit zu bekannten, an STL-Stoffwechselwegen beteiligten P450-Enzymen gesucht. Mithilfe von 3´- und 5´-RACE-PCR wurden die offenen Leserahmen der Kandidaten aufgeklärt. Die anhand dieses Verfahrens ausgewählten, sowie weitere bereits beschriebene P450 Kandidaten Sequenzen wurden in Hefe-Vektoren transformiert und in Kombination mit verschiedenen Substrat-Vektoren exprimiert. Ein Hochdurchsatz Mikro-Ansatz wurde entwickelt, der die Expression und Analyse vieler Hefe Stämme im selben Zeitraum ermöglichte. Für die transiente Expression in N. benthamiana wurden die entsprechenden Gene bekannter und putativer Enzyme über Agrobakterien-Transformation in Tabak-Pflanzen eingebracht. Beim in planta Expressions-System wurde der komplette STL Stoffwechselweg der Sonnenblume bis zum Costunolid de novo in einem Schritt-für-Schritt Ansatz rekonstruiert. Die bereits bekannte Additionsreaktion von α-Methylen-γ-Lacton STL mit der Thiol-Gruppe von Cystein und Glutathion konnte bei allen untersuchten STL beobachtet werden. Als Referenz wurden STL-Addukte chemisch synthetisiert und mit den in planta erzeugten Produkten abgeglichen. Die Charakterisierung von Enzymen erfolgte somit in zwei unterschiedlichen in vivo Expressions-Systemen in Hefe (Saccharomyces cervisiae) und Tabak (Nicotiana benthamiana). Für den vorliegenden Stoffwechselweg wurden eine Reihe Unterschiede und Charakteristika der beiden Expressionssysteme herausgearbeitet. Kandidat M4 zeigte in Hefe ein unerwartetes Produkt (Farnesyl-δ-Lacton) und führte in Kombination mit HaG8H zur Produktion von Costunolid. Im pflanzlichen Expessions-System wurde Germacren A Säure von M4 auch in Abwesenheit von HaG8H zu Costunolid überführt. In beiden Fällen trug Kandidat M4 zur Synthese von Costunolid bei, und kann somit als Helianthus annuus Costunolid Synthase (HaCOS) bezeichnet werden, allerdings sollte der zu Grunde liegende Reaktionsmechanismus noch weiter aufgeklärt werden.
Die Helianthus annuus Costunolid 14-Hydroxylase HaC14H (Kandidat M33) wurde in Hefe und Tabak charakterisiert, eine Einteilung in die Unterfamilie CYP71CB, zusammen mit TP8878, der Tanacetum parthenium Costunolid/Parthenoild 3β-Hydroxylase wird vorgeschlagen. Des Weiteren konnte gezeigt werden, dass in planta das Hauptprodukt der HaG8H höchstwahrscheinlich als Inunolid vorliegt, was den Biosyntheseweg hin zu 8-Epixanthatin und Tomentosin einleiten würde. Ein weiteres Enzym, Kandidat S2 aus der Arbeit von Ikezawa et al. (2011), konnte in planta, nicht aber in Hefe, 8β-Hydroxy-Germacren A Säure in 8β-Hydroxy-Costunolid/Eupatolid und mehrere Nebenprodukte umsetzen, dementsprechend wurde der Name Helianthus annuus Eupatolid Synthase, HaES vorgeschlagen. HaES besitzt 47 % Aminosäure Identität zur Parthenolid-Synthase von T. parthenium TpPTS, es wird eine Einteilung in eine eigene CYP71 Unterfamilie vorgeschlagen. Zwei alternative metabolische Routen führten bei der Expression in planta zur Bildung von 8β-Hydroxy-Costunolid, die zugrundeliegenden Mechanismen wurden diskutiert. Analysen der Expressionsmuster der am STL-Biosyntheseweg beteiligten Enzyme zeigen, dass die Gene auf den STL Synthesewegen auch in den inneren Geweben exprimiert sind und deren Expression in Blattprimordien parallel zur KDH Entwicklung und STL Produktion koordiniert hochreguliert wird. HaC14H lag in einer Region des Genoms in der Nähe einer Reihe weiterer P450 Enzymkandidaten, die sich in dieselbe Unterfamilie eingruppieren lassen und ebenfalls in den KDH exprimiert sind, weshalb eine Beteiligung von P450 Enzymen aus dieser Gruppe an weiteren Reaktionsschritten hin zu den komlpexeren STL der KDH plausibel wäre.
 
Kurzfassung auf Englisch: In the present work additional steps towards the elucidation of the biosynthetic pathway of H. annuus sesquiterpene lactones (STL) were achieved. Firstly candidate sequences were retrieved from a transcriptome database by filtering according to expression pattern and similarity to P450 enzymes known to participate in STL biosynthetic pathways. Open reading frames (ORFs) were obtained using 3´-and 5´-RACE-PCR. Previously described and newly identified candidate genes were then transformed in yeast vectors and expressed in combination with different substrate vectors. A high throughput micro approach was developed that allowed the expression and analysis of many yeast strains at the same time. For the transient expression in N. benthamiana the genes of known and putative enzymes were introduced via Agrobacterium mediated transformation. Using the in planta expression system the complete STL pathway of sunflower to costunolide was reconstructed de novo in a step-by-step approach. Previously described Michael-addition reactions of α-methylene-γ-lactone type STL to the thiol group of cysteine or glutathione in tobacco expression systems could be observed for all STL investigated. Chemically synthesized STL adducts were used as reference for the identification of in planta produced STL adducts. Enzyme characterization was conducted in two different in vivo expression systems, in yeast (Saccharomyces cervisiae) and tobacco (Nicotiana benthamiana). For the investigated biosynthetic pathway, differences between these two expression systems were discussed. Candidate gene M4 showed an unexpected product in yeast (farnesyl-δ-lactone) and led in combination with HaG8H to the production of costunolide. In the plant expression system, germacrene A acid was converted to costunolide by M4 in the absence of HaG8H. In both cases, M4 was involved in the synthesis of costunolide and should therefore be assigned Helianthus annuus costunolide synthase the underlying reaction mechanism should however be investigated more thoroughly.
Helianthus annuus costunolide 14-hydroxylase HaC14H (candidate M33) was characterized in yeast and tobacco. A classification into subfamily CYP71CB, together with Tp8878 the Tanacetum parthenium costunolide/parthenolide 3β-hydroxylase is proposed. It was shown that in planta the main product of HaG8H exists most likely as inunolide, which would be the entry point for the biosynthesis of 8-epixanthatine and tomentosine. Candidate S2 from Ikezawa et al. (2011) was found to convert 8β-hydroxy-germacrene A acid to 8β-hydroxy-costunolide (eupatolide) in tobacco, but not in yeast, producing several byproducts. The name Helianthus annuus eupatolide synthase HaES is proposed accordingly. HaES has 47 % amino acid identity to the parthenolide synthase from T. parthenium (TpPTS). A classification into a new CYP71 subfamily is proposed. Two alternative metabolic routes led to 8β-hydroxy-costunolide in the expression studies in tobacco, the underlying mechanisms are discussed. Enzymes involved in STL biosynthesis were expressed in inner tissues of young Helianthus annuus plants; the induction of expression of STL biosynthesis enzymes in leaf primordia correlates with the development of capitate glandular trichomes (CGT) and STL synthesis. HaC14H was found in a chromosomal region in proximity to several P450 enzyme candidates, that share the same subfamily and the expression in capitate glandular trichomes (CGT). Therefore involvement of these enzymes in later steps of the biosynthesis of the elaborate STL structures found in CGT is likely.
